    摘要:

    目的观察不同来源的神经干细胞(NSCs)上清液对骨髓巨噬细胞的增殖、吞噬及分泌细胞因子的影响,为探讨NSCs治疗疾病提供新的思路。方法采用原代培养及胚胎干细胞(ESCs)来源 NSCs的上清液处理骨髓来源的巨噬细胞,通过SRB比色法检测细胞增殖情况;油红及免疫荧光染色观察巨噬细胞的吞噬能力; ELISA法检测各组巨噬细胞上清液中TNF-α、IL-6及IL-10的表达情况。结果骨髓来源细胞在含L929上清液的培养液中培养7 d可获得良好的巨噬细胞。 NSCs培养基组、 NSCs上清组和ESC-NSCs上清组巨噬细胞增殖率分别为(100±1.2)%,(135±2.1)%和(132±3.4)%,油红阳性细胞百分率分别为(50.7±4.5)%,(92.2±3.7)%和(89.5±4.3)%。相比NSCs培养基组, NSCs上清组和 ESC-NSCs上清组的IL-6、IL-10含量增高,而TNF-α含量下降(P< 0.05)。结论原代培养的NSCs、 ESC来源的NSCs上清液均可促进骨髓巨噬细胞增殖,增强其吞噬能力。

    Abstract:

    Objective To explore the influence of neural stem cells(NSCs)supernatants on bone mar-row derived-macrophage,in order to provide new ideas for therapy. Methods Mouse bone marrow macrophage were treated with NSCs supernatant for 3 d. SRB assay was used to detect the proliferation of macrophage. Thephagocytosis of macrophage were detected by oil red staining after adding myelin for 24 h. Meanwhile,the expression of TNF-α,IL-6 and IL-10 in the macrophage supernatant were detected by ELISA kit. Results Macrophagewere obtained after bone marrow-derived cells cultured in the medium containing L929 supernatant for 7 d. Re-spectively,macrophage proliferation rate of NSCs medium group,NSCs supernatant and ESC-NSCs superatant group was(100±1.2)%,(135±2.1)% and (132±3.4)%. Oil red percentage of positive cells was (50.7±4.5)%,(92.2±3.7)% and (89.5±4.3)% respectively. Compared to NSCs medium group,the levels of IL-6 and IL-10 in macrophage treated with NSCs and ESC-NSCs supernatant significantly increased,and the expression of TNF-α significantly decreased (P< 0.05). Conclusions Both primary NSCs supernatant and ESC-derivedNSCs supernatant could promote the proliferation and phagocytosis of bone marrow macrophage.
