Abstract:Objective To investigate the relationship between Notch signaling pathway and epithelial mesenchymal transition in glioma cells. Methods Immunomagnetic bead was used to isolate brain glioma stem cells from human glioma tissue. The glioma stem cells were cultured in vitro. Immunofluorescence assay was used to identify the cells. RNA interference retroviral vector of Notch-1 gene( pSiRNA-Notch-1) was constructed. The experiment contained three groups: blank control group( non-transfected plasmid), negative control group( transfected with blank plasmid) and interference group( transfected with pSiRNA-Notch-1). The proliferation and differentiation of three groups were observed. The expression of mRNA and protein of Notch-1 and Hes-1 was detected by RT-PCR and Western blot. Results The cell proliferation and cell differentiation absorbance( OD) of interference group after 7d were( 1.332±0.614) and( 1.203±0.783) respectively, which were significantly lower than that of blank control group and negative control group( P<0.05). The relative expression of mRNA and protein of Notch-1 and Hes-1 in interference group after 3 d proliferation was( 0.189±0.021) and (0.301±0.121),( 0.422±0.022) and( 0.091±0.032), which was significantly lower than that in blank control group and negative control group( P<0.05). The relative expression of mRNA and protein of Notch-1 and Hes- 1 in interference group after 3 d differentiation was( 0.253±0.071) and( 0.192±0.043),( 0.178±0.022) and (0.101±0.012), which was significantly lower than that in blank control group and negative control group( P<0.05). Conclusions Notch signaling pathway plays an important role in proliferation and differentiation of glioma cells. Notch-1 and Hes-1 may be involved in the regulation of proliferation and differentiation, which needs further study.

Abstract:Objective To explore the changes and clinical signification of von Willebrand Fator Antigen( vWF:Ag), D-dime(r DD) and Fibrinogen Degradation Products( FDP) in acute cerebral infarction patients. Methods Levels of vWF:Ag, DD and FDP were measured by immune nephelometry. The differences of vWF:Ag, DD and FDP were analyzed between two groups. And the levels of vWF:Ag, DD and FDP were followed up for one year after the event. Results Totals of 94 patients with acute cerebral infarction were included [56(59.6%) males and 38(40.4%) females]. 120 healthy controls were included in the control group[68(56.7%) males and 52(43.3%) females]. Levels of vWF:Ag and DD in patients group were higher than that in control group(P< 0.01). The logistic regression analysis showed that increasing of vWF:Ag and DD were risk factors of acute cerebral infarction. Levels of vWF:Ag and DD were positive correlated with score of NIHSS(r=0.625,0.510; P< 0.01). Level of vWF:Ag was positive correlated with DD(r=0.688,P< 0.01). Levels of vWF:Ag, DD and FDP in patients with poor prognosis were higher than that in patients with good prognosis(P < 0.05). Conclusions Levels of vWF:Ag and DD are signifiantly increased in acute cerebral infarction patients, and positive correlated with NIHSS score. Detection of vWF:Ag and DD in acute cerebral infarction patients is contributed to the judgment of severity and prognosis of the disease.

Abstract:Objective To investigate the changes of Genomic DNA methylation from whole blood samples in bipolar disorder patients. Methods Genome-wide DNA methylation levels in whole blood obtained from 10 bipolar disorder patients and 5 healthy controls were assayed by Infinium Human Methylation450 BeadChip (Illumina). The association of DNA methylation with bipolar disorder was analyzed by IMA of R project. Gene ontology and Pathway were analyzed by GCBI. Results Compared to healthy controls, DNA methylations of SLC6A3, ARNTL, MAGI2, FAM111A, ZNF578 and FAM196A genes were significantly upregulated, while DNA methylations of SMPD1, ZEB2, KCNQ5, FAM41C and RGS18 genes were significantly downregulated in bipolar disorder patients. Conclusions DNA methylation may be associated with bipolar disorder.

Abstract:Objective To explore the characteristics of cognitive impairment and sleep disorders in patients with severe stenosis or occlusion of internal carotid artery. Methods Forty patients( case group) with severe internal carotid artery stenosis or occlusion were selected from January 2015 to June 2016 in Jiangmen City Central Hospital. Eighty patients with no stenosis of internal carotid artery were enrolled as control group. The Montreal Cognitive Assessment scale( MoCA), Mini Mental State Examination( MMSE) and Pittsburgh Sleep Quality Index scale( PSQI) were assessed to measure the cognitive function and sleep quality in two groups. Results Scores of visual space structure and execution, attention, memory, language, abstract thinking, name, time and place of directional function and MoCA score were lower in case group than that in control group (P< 0.05). Scores of orientation ability, language, memory, memory, calculation ability and MMSE scores were lower in case group than that in control group( P < 0.05). Scores of sleep time, sleep efficiency, sleep time, sleep quality, daytime dysfunction and hypnotic drugs, PSQI scores were higher in case group than that in control group( P < 0.05). Conclusions Patients with severe stenosis or occlusion of internal carotid artery have cognitive impairment and decreased sleep quality.

Abstract:Objective To investigate the compliance and its influencing factors of patients with schizophrenia. Methods The psychological data of antipsychotic drugs were evaluated in the form of semi-structured interview after 3 weeks of hospitalization by Rating of Medication Influence Scale( ROMI). The relevance of demographic data and clinical data were evaluated, and the impact of ROMI factors on compliance was predicted. Results The demographic factors( age, education, working status, supervision, payment) and clinical factors( type of medication, duration of disease, number of hospitalizations) were not associated with ROMI score( P>0.05). While drug side effects and ROMI non-compliance scores were positively correlated( P<0.05). Patients followed the instructions of experts, felt the benefits of drugs on daily life, feared of recurrence, family members or friends had a positive opinion on the use of drugs play an important role among the factors( P<0.01). Patients considered that the side effects of drugs were more moderate, the drug could help achieve personal goals and had a good relationship with medical staff play an important role in impact factors(P < 0.01). Conclusions Drug side effects are common factors that affect medication compliance. Following the instructions of the expert, feel the benefits of the drug to the daily life, fear of recurrence, family or friends have a positive opinion on taking the drug are more greater factors which affect medication compliance.

Abstract:Objective To intellectual development of urban and rural children in Xiamen through wechsler intelligence scales of different age. Methods A total of 233 children aged 6-16 years from urban and rural areas in Xiamen were enrolled in this study by random stratified sampling method. C-WISC and WISC- Ⅳ were used to measure the IQ. Results In urban children, the coding and digit span score in WISC-Ⅳ was significantly lower than the corresponding score in the C-WISC( P<0.05); the block design score in WISC-Ⅳ was significantly lower than the corresponding score in C-WISC(P< 0.05); the verbal intelligence quotient score in WISC-Ⅳ was significantly higher than the corresponding score in C-WISC(P< 0.05); the similarities score in WISC- Ⅳ was significantly higher than the corresponding score in C-WISC(P < 0.05). And there were no significant differences in the scores of comprehension, perception reasoning, total intelligence quotient between the two methods( P>0.05). However, in rural children, each score in WISC-Ⅳ were significantly lower than the corresponding score in C-WISC( P<0.05). Conclusions There is difference on intellectual development between urban and rural children in Xiamen. Urban children’s intellectual development is balanced, but rural children’s intellectual development is lagging behind.

Abstract:Objective To study the efficacy of improved Naikan therapy combined with durgs on adolescents with anxiety. Methods Totals of 61 adolescents with anxiety who were treated in Psychological Clinic, People’s Hospital of Zhongshan City were randomly divided into study group( n=32) and control group (n=29). Patients in study group were treated with sertraline combined with improved Naikan therapy, while those in control group were treated with sertraline combined with general psychological counseling therapy. Hamilton Anxiety Rating Scale( HAMA), Self-rating Anxiety Scale( SAS) and Diagnosis Scale of Social Adaptability were measured before and after 1, 4 and 8 weeks’ treatment on all patients. Results Scores of HAMA and SAS in both groups were significantly lower after 1, 4 and 8 weeks’ treatment( P< 0.05). Scores of HAMA and SAS after 1, 4 and 8 weeks’ treatment in study group were significantly lower than that in control group( P<0.05). Scores of Diagnosis Scale of Social Adaptability in both groups were significantly higher after 1, 4 and 8 weeks’ treatment( P<0.05). Scores of Diagnosis Scale of Social Adaptability after 1, 4 and 8 weeks’ treatment in study group were significantly higher than that in control group( P< 0.05). Conclusions The efficacy of improved Naikan therapy combined with drugs is obvious.

Abstract:Objective To investigate the correlation between levels of interleukin-6( IL-6), interleukin- 8( IL-8) and tumor necrosis factor-α( TNF-α) in peripheral blood and cognitive function in patients with Alzheimer disease( AD). Methods Ninety patients with Alzheimer disease treated in Yanzhou Hospital of Affiliated Hospital of Ji’ning Medical College from January 2015 to January 2016 were selected as the study group. Ninety healthy subjects who were examined at the same period in our hospital were selected as the control group. IL-8 and TNF-α levels in peripheral blood were detected by enzyme-linked immunosorbent assay( ELISA). The cognitive function and ability of daily living were assessed by Mini-Mental State Examination (MMSE) and Clinical Dementia Rating Scale( CDR). The differences of inflammatory factors between two groups were compared. And the correlation between the expression of inflammatory factors and cognitive function was analyzed. Results The levels of IL-6, IL-8 and TNF-α in peripheral blood in the study group were significantly higher than that in the control group[ (128.64±10.28)pg/ml vs( 103.59±8.72) pg/ml,( 134.32±9.67)pg/ml vs( 101.45±7.32) pg/ml,( 221.39±23.54)pg/ml vs( 109.68±18.76) pg/ml, P< 0.05]. Education was a protective factor of cognitive function in AD patients, while the levels of IL-6, IL-8 and TNF-α were risk factors (P< 0.05). IL-6 and TNF-α were negatively correlated with MMSE scores in all subjects(r=-0.314, -0.079; P< 0.05). But no correlation was found between IL-8 and MMSE scores. Conclusions The levels of IL-6 and TNF-α in peripheral blood were correlated with cognitive function in patients with mild AD.

Abstract:Objective To study the protective effect of bilobalide on cerebral ischemia reperfusion injury under hyperglycemia in rats and explore the possible mechanisms. Methods Rats were given 50% glucose( 6 ml/kg) by intraperitoneal injection to get acute hyperglycemia model. Cerebral ischemia reperfusion model was made by intraluminal monofilament. 40 rats were randomly divided into hyperglycemia+sham operation group( sham group, n=8), hyperglycemia+MCAO group( model group, n=8), hyperglycemia+MCAO+bilobalide group( bilobalide groups, n=24). Bilobalide groups were then divided into three dose groups( 2.5, 5 and 10 mg/kg, n=8). The bilobalide groups were given intraperitoneal injection for 3 d and 1 h before cerebral ischemia. After blocking middle artery for 2 h and reperfusion for 24 h, neurological deficit scores, cerebral infarction volume, brain water content, AQP4 mRNA, SOD activity, MDA content, NE, DA and 5-HT expression were detected. Results Rats in bilobalide( 5, 10 mg/kg) group showed significantly relieved neurological deficit scores, reduced cerebral infarction volume, decreased brain water content, down-regulated AQP4 mRNA expression, up-regulated SOD activity, reduced MDA content and improved content of NE, DA and 5-HT, compared to those in model group(P< 0.05). Conclusions Bilobalide could relieve cerebral ischemia reperfusion injury of cerebral ischemia reperfusion rats under acute hyperglycemia.

Abstract:Objective To investigate the correlation between levels of serum angiopoietin-1( Ang- 1) and the severity of acute cerebral infarction, in order to provide evidences for early clinical evaluation of acute cerebral infarction. Methods Patients with acute cerebral infarction in the Ninth Hospital of Xi’an City were enrolled as the case group. In the corresponding period, healthy subjects in physical examination center were enrolled as the control group. The general clinical data of all subjects were collected. The levels of serum Ang-1 were measured in all subjects. The patients were divided into three groups according to NIHSS score or maximum infarct diameter in MRI respectively. The difference of clinical data between the case group and control group were analysed. Logistic regression analysis was used to analyse the risk factors. ROC curve analysis was used to evaluate the diagnostic value of serum Ang-1. Spearman correlation analysis was used to analyze the correlation. Results The percentages of smoking, hypertension, diabetes mellitus and atrial fibrillation were significantly increased in the case group, while the levels of serum Ang-1 were significantly decreased (P < 0.05). Logistic regression analysis showed that diabetes mellitus and the levels of serum Ang-1 were risk factors of cerebral infarction( P<0.05). ROC curve analysis showed that the area under the ROC curve was 0.791 (95%CI=0.751-0.830) about the diagnosis of acute cerebral infarction with the level of serum Ang-1. Significant differences were found between the levels of serum Ang-1 in mild, moderate and severe NIHSS group( P<0.01). Significant differences were found between the levels of serum Ang-1 in large infarction, middle infarction and small infarction group(P< 0.01). Spearman correlation analysis showed that NIHSS score and maximal diameter of infarction in MRI were negatively correlated with the serum level of Ang-1( r= -0.498, -0.459;P<0.01) in acute cerebral infarction patients. Conclusions The level of serum Ang-1 in patients with acute cerebral infarction decreases, which is closely related to the severity of cerebral infarction.

Abstract:目的 探究问题导向干预模式护理对帕金森病老年患者心理、睡眠与生活质量的影响。 方法 回顾性分析2014年1月~2015年1月我院收治的100例老年帕金森病患者的临床资料,作为对照 组,实施常规护理;2015年1月~2016年1月收治的老年帕金森病患者100例作为观察组,实施常规护理+ 问题导向干预模式护理。比较两组患者干预前后的心理、睡眠与生活质量情况。结果 观察组干预护理 频率低于对照组,组间比较差异具有统计学意义(P< 0.01);与护理前相比,两组患者护理后的PSIQ总分、 SAS评分、SDS评分均降低,生活质量得分均升高,护理前后比较差异有统计学意义(P< 0.01);且以观察 组护理后各项评分优于对照组,组间比较差异有统计学意义(P < 0.01)。结论 应用问题导向干预模式 对帕金森病老年患者进行护理的效果显著,心理状态与睡眠质量均得到有效改善,生活质量提高,值得 在临床上推广应用。

Abstract:认知功能障碍是精神分裂症患者的核心症状之一,与患者的预后和社会功能关系密切,使 用抗精神病药物改善精神分裂症患者的认知损害效果尚不理想。重复经颅磁刺激(rTMS)作为一种将脉 冲磁场作用于大脑皮质并产生感应电流,进而影响脑内神经电活动和基础代谢的电生理技术,目前已被 用于精神分裂症患者的临床治疗中。本文综述了rTMS改善认知功能的可能机制,对rTMS治疗精神分裂 症患者认知功能障碍的研究进展以及安全性等进行阐述。
